Samus, I feel, should be a lot like Batman: someone who's been so affected by loss and grief that it disconnects her from the rest of society somewhat. This is someone who lost their parents at an early age, who was taken in by an alien race, who also deserted her eventually. This is a fundamentally lonely person, one who prefers to keep to themselves almost all of the time.
And yet despite all the loneliness and isolation, she should be someone stable and socially adept enough to be cool and confident in social situations, she just carries herself with a quiet, reserved demeanor. But underneath that introverted exterior is a woman who defies any one sort of grouping, aside from being a badass space bounty hunter warrior. She should be compassionate and loving, particularly towards civilians, children, animals, fallen comrades, but stern and determined to take down the Space Pirate menace and any other sort of evil in the galaxy, even having a sort of hatred for that sort of thing.
But I feel like she should also have a sort of dry, sarcastic sense of humor that she'd show every once in a while, particularly to her comrades, like Adam Malkovich or Anthony Higgs (who should also be written completely differently than how they were in Other M).
I think she should also be somewhat of a biological scientist as well, also somewhat of a xeno-archaeologist. Remember, this is someone who scans and catalogs everything she sees on her adventures. She should absolutely take interest in the study of living creatures, especially alien, foreign ones.
And she should, every so often, show glimpses of a warm, even somewhat happy core to her, someone who, despite everything, is happy that she's alive and happy with everything she's had in life. Even if we don't see that side of her all the time, it should still be there deep down.
