And Rule 63 for the most part actually seems to have become popular because of either straight males with a feminization fetish, or straight males who are turned on by the novelty of a familiar, aesthetically appealing design rendered in a form that is to them fappable.
So if anything, Rule 63 is for straight males, not trans.
The real trans bait is just a good-looking, likable character that can be thought of as an ideal, a goal to become like. For instance, Danny Fenton. There's a ton of people in the surviving fandom that like to think of him as trans. You can never really predict if a character will be trans bait until you see their reception on Twitter. The main thing seems to be not being too feminine or too masculine, but that applies to a lot of characters.
Giovanni from that web series Epithet Erased was massive trans bait, but the creator actually stepped in and said that that was a no-go, since Giovanni was an avatar of an IRL person, and that person isn't trans
