This issue is "The Uncanny Life of Moira X (pronounced Ten)."

Simple summary: There are no X-Men in this issue. It picks up from the X0 scene from Powers of X #1. Hickman gives us one extended flashback scene (in the form of TEN) that changes everything about the history of the X-Men, and he explains why Moira, not Xavier, is the most important character in the history of the X-Men.

As it turns out, Moira is a mutant. Her power is that she lives her life and then is reborn to live her life again with all of the memories and experience of her previous lives; and she has ten of them. The current timeline that everything has ever happened in is her tenth life, and she's spent her previous nine lives trying to solve "the mutant problem." House of X is the culmination of what she's learned.

The biggest retcon is that in this tenth life, when Moira meets Xavier, she shares her all of her previous lives with him, so House of X is what Xavier (along with Moira) has been working towards this entire time.
