Lips can be defined by the negative space between them, such as a shadow or an open mouth. This is often more defining than the positive shapes of the lips.
The transition between the two types of skin is subtle. You also have to preserve three dimenshionality when you draw the darker type of skin. I think of the upper lip as being three shapes and the lower as two.
