Histology: Cell orientation in tissue samples

Do cells of the same type ever end up in a mix of both A (flat) and B (upright) orientations in the same tissue sample? Or are there specific steps in the preparation of tissues that positions all of its elements in the same "flat" orientation?

Also, are there ever times where either orientation, or a mix of the two, can throw off an observer's ability to understand what type of tissue they are looking at?
