The author suggests hypothetical expression of reverse transcriptase from dormant LINE-1 elements in the human genome, but despite his experimenting is unable to measure this actually happening. He makes a series of conjectures which pose a purely hypothetical pathway for SARS-CoV-2 to integrate into the human genome, but is unable to support it.
If this particular virus could integrate into our genome via this proposed mechanism, then why do not all viruses do so? I will grant that if you have HIV then it is possible due to that virus coding for presence of RT, but other than that it is pure hypothesis.

Tracking down the agent of RT-activity is also difficult here. ut what we know is that once there is cellular stress there are endogenous retroviruses roaming around, thus RT-activity. Combine LNP-cytotoxicity and a huge amount of exogenous RNA inside a cell and you got yourself some genome-integration.

And still, it's a short, pre-print paper. Who knows what they know more know and try to publish.

Why does this matter?
Virus inject their RNA into a cell and force it to make more viruses. Eventually the cell makes so many viruses, it either bursts, or it dies from not doing other critical processes and bursts, releasing the viruses.

Even if the viral RNA integrates to the DNA in the cell, that cell shouldn't reproduce, right? If the cell doesn't reproduce, then the chimeric DNA dies with the cell.

Not a filthy biologist, so maybe I have something wrong.

Of course things move from the extracellular interstitium into the blood stream. It's just very slow and regulated by the epithelium around a vessel. Vaccines do not contain things that readily cross that barrier. Instead, they contain things that are readily taken up by dendritic cells and muscle cells. The fluid part of vaccines do diffuse, but it's just saline anyway. The molecules within do not diffuse readily, and never get close to the bloodstream.

LNPs do absolutely, and easily cross membranes (btw, vessel have endothelium, not epithelium, retard). The chemical nature and small size of those nano particles gives it characteristics that permit easy diffusion through tissue. Like steroids, or benzene.

Lastly, to think there are two phases in a vaccine (solid and fluid) is absolutely moronic. Especially for LNP solutions thats not true

Also someone with healthy hemostasis won't throw venous thromboses just because a vein is punctured. The coagulation cascade contains a lot of anti-thrombotic signals that kick in immediately to prevent over-coagulation. People get holes in veins all the time.

The spike proteins, regardless of source, are inflammatory. If your body continues making them, your body will be in a constant state of inflammation. The theory has been that the mRNA would break down and the cells doing its bidding would die out like cells do. But if they're propagating with their protein spike producing ability intact, that's a problem.

The cells that make up the endothelium are squamous epithelial cells, my dude.

LNPs are precisely the molecules that are very readily taken up by DCs, and don't make it to the blood stream. They're also cleared pretty quickly, as would any mRNA contained within. Their role is solely to deliver the mRNA to professional APCs. Thinking that getting them injected into the muscle and they make it to the bloodstream is simply wrong, and you know that.

Even if this true

1) you'd have more integration events over the course of a natural infection compared to a vaccine and

2) cell lineages experiencing reverse transcription/integration would be restricted to terminally differentiated cells with relatively short half lifes in the case this is occurring with the vaccine as well ( which it probably isn't)

And that would be in the worst case scenario, which is unlikely anyways. We don't really know what is happening in vivo when patients are persistently expressing viral gene products (this is only a small subset of covid patients as well), this is all hypothesis and supposition with little empirical evidence.

I've looked at a couple years old study where they measured luciferase-activity througout the body after delivering luciferase gene via PEGylated LNPs (what we have in the rona vaccines). It showed that in mice half life after intramuscluar injection is many days (meaning it will be around for many weeks, in humans, many months), and mostly concentrated at liver and lungs. EMA report shows destruction of portal liver tisse, hints to the same thing.

The study is stating a fact: cured people still test positive. They try to find an explanation, but even if it is wrong or partially wrong in the mechanisms involved, it is hard to find another explanation than reverse transcriptase.
