You cannot manipulate the tunnelling amplitude; it is purely stochastic. I'm not sure what conditions you have in mind, but say you're talking about an alpha particle tunnelling out of a nucleus, the probability is constant over time, it isn't "zero until the conditions are met then it's becomes 100%", it's somewhere between 0 and 1 until you measure it, at which point it becomes certain. That isn't a condition being met and making it happen, that's it happening by random chance and you OBSERVING it having happened.
