Are some drugs like LSD and THC not dose dependent, while drugs like alcohol and mdma are HIGHLY dose dependent?

How exactly does LSD enter and travel through the body, is there any info on that? I've looked up several studies and articles and no one really get's into this, they all talk about how it acts with the receptor and somethings about the pharmacokinetics

There ARE drugs like Gabapentin with weird dose-effect dynamics but that can normally be attributed to drug metabolism rather than direct mechanism. Generally, if you had something that performed a single, direct task, then got metabolized via a single pathway, there will be a fairly linear relationship between dosage and effect. At least until its target binding site or something in its metabolic pathway becomes saturated. The ambiguous thing here is that nothing behaves cleanly like that. There are almost always multiple avenues of metabolism and exhibit their subjective effects (assuming we're talking about fun drugs here) here through multiple layers of cascading effects of drug binding.

no i get it but that doesnt answer why some drugs basically ignore how heavy a person is, afaik it had something to do with how easily it passes the BBB but I dont remember and this is why I opened this thread

so if a person is bigger/heavier, blood concentrations of any drug will obviously be lower with the same dose. Hmm, I'm just spitballing but since you mentioned the BBB I'm inclined to say that some drugs can be taken in (to wherever their target may be) so efficiently that being forced in via concentration gradient isn't as necessary... so regardless of the blood concentration a similar amount of the drug may actually affect its target site.

Gabapentin (this is more related than I thought) has to be actively moved across the BBB through a pathway that saturates very easily. Because of this, dose-response starts out linear but begins to level out at higher doses. So you end up with diminishing returns taking more of it... and some of it ends up being metabolized into something inactive before it can do anything.

Well the specific pathway will be completely different depending on what you're looking at. Some drugs closely resemble endogenous substances so they already have robust pathways for uptake and metabolism, some drugs are just weird enough to hijack parts of these pathways, some can bind allosterically in places nothing else does naturally. There are surely general trends in drug behavior but it really does depend on the specific drug. In biochem, even tiny differences can change everything.

How is THC not dose dependent? Sure its LD50 approaches infinity (or at least high enough you cannot reach it via inhalation) but the more you smoke in a sitting the more stoned you get.

because it is visual receptors in the brain being inhibited and folded into themselves
even then, LSD is absolutely dose dependant, if you take 1/10th of a regular dose, you'll get a far smaller effect, if you take an entire sheet's worth of LSD then you'll pass out, vomit, mild seizures (according to "overdose" tales)
