Whether or not humanity develops into an interstellar civilization is going to depend entirely on the FTL question - are any of the "loopholes" for getting around the lightspeed barrier possible from a physics standpoint and feasible from an engineering standpoint. If so, then interstellar civilization is possible, if not, it's not.
Don't misunderstand - I don't doubt that we'll still colonize worlds in other star systems, but if FTL isn't a thing, it's going to look VERY different compared to your United Federations, your Terran Federations, your Systems Alliances, your Twelve Colonies, etc. Instead human colonies in different systems will be extremely isolated. Interstellar communication will be minimal, interstellar trade will be almost non-existent, etc. Within a few dozen or hundred generations as humans adapt to their new homes we may even start to see physiological divergence.
