If not from amino acid chains then how about from a pig/cattle pancreas? What is the process and what equipment would it require? I have a family member with type 1 diabetes and have concerns about civil unrest causing extended disruptions in the supply chain.

IIRC Insulin is synthesized by copying the gene into yeast which can quickly multiply and produce it in bulk. Prior to that, they would harvest it from pig pancreases; While this was better than dying, there was a chance of the user falling ill due to allogenic complications (similar to that encountered in transplants).

While I'd say the latter is more feasible than the former from a diy perspective, It's still challenging. You'd be better off stockpiling (I believe that insulin can be refrigerated for longer at extremely low temperatures).

This only yields results for people who have created an "artificial pancreas" using a CGM, insulin pump and bootleg software to deliver a constant insulin flow. I'm looking to synthesize insulin itself.

Look up the open insulin project and see if they are already in your state or nearby. If not, see if any universities or county governments near you would be interested in a partnership.

A closed loop system is able to alter itself based on feeback it generates (compared to an open loop system where an actor takes the feedback and alters the system accordingly). I don't know if you're confusing it for other potential diabetes treatments but closed loop insulin pumps are not self-sustaining.
