Order the plasmid you want/need and do some optimization of your transfections.
Once you figured out how to do that do a bulk transfection, select with marker and resistance gene of your choice, pick a cell that survived the treatment and grow a new culture from that.
Run WB and qRT-PCR to confirm your target.
