Although too be honest it's a real bitch to work with. At least with folded proteins and stuff I can easily keep it in solution and not stress about it turning into an unworkable gel if I get it too concentrated before I make all the samples I need. Like, the gel is part of what I'm studying, but jesus christ there is nothing as frustrating as getting everything ready, finding all the right concentrations, then having it get pissy right before you were about to do something and it just decides to crash+gel.

Yes, but you need physical intution. Some form of intimate familiarity with biological systems, otherwise you will propose ludicrously overcomplicated abstract theory with no predictive power. This is fairly common for mathematicians and physicists to fall into when they dabble in bio. But, things are improving considerably now that there are larger datasets available and more institutional support for biophysics. Today having mathematical competence would put you in the top 99th percentile of biologists purely because most of them can barely compute a basic trig integral without a calculator.

I think it aggregates due to the up tick in beta-sheets I read from my FTIR data. Really the aggregation shouldn't be a huge problem, just toss it in some urea and all the beta sheets break apart.

But the problem is that it happens right before I'm about to study the high concentration gel structures. So it's very very frustrating. I was debating just adding a crowder in to simulate a higher concentration to see if I could induce the conformation change I'm going after. But my boss would force me to use all of our crowders and I'd rather die then waste a year and a half doing that.

Luckily we can get 30 mg/L and the bioreactor I set up lets me pull 16 L in a day if I'm being slick.

Any advice?

Have you done DLS or AUC experiments to see you are getting multiple diffusion coefficients?
If you are getting a lot of aggregation the usual advice is add glycerol (5-10% mg/mg) because that can stabilize the backbone H-bonds if they are solvent exposed to prevent higher order oligomeric structures from arising. I have had an issue with a protein where it crashes out of solution instantly when the conc. is too high and found another issue is salt concentration.

If your salt is too low (50-100mM) that can cause the crashing out. I used 250mM NaCl and I can go up to about 100mg/mL on this protein.

This protein is very tricky, I am looking into AUC experiments right now but DLS is pretty much a non-starter due to some of its physical properties. I will look into the glycerol thing, although in general anything that alters the material properties is suspect.

We did try a range of salts and the like, it worked okay but had a massive effect on gel strength which makes me leery of using it in my studies.
