>>11679688use the agar strain isolation technique. To get good, virulent mycellium, you want a monoculture, or at least something close to a monoculture. Also, strain isolation on agar makes it very easy to propagate your myc. Ideally, you should build yourself a laminar flow hood, but a plastic/rubber glovebox works in a pinch. Make sure never to expose the inside of your GB to a room, try to keep the room very clean, and spray the inside of the GB with dilute bleach before you start working with your petris. The bleach is antibacterial/antifungal, but the main point is to keep the interior of the GB coated in water droplets, because all the floating particulate will generally stick to the moist walls, and that will keep things from going contam. Get a minifridge for storing your petris. Start with this step. Order some spores from Free Spore Ring Earth and start isolating your strain. Once you've got an okay virulent strain, propagate it to 15-20 petris and save them in your minifridge. This will keep you busy for a couple of months.
