If you mean in the "Grand Scheme" of existence, would everything being a simulation change the overall motive of scientific progress. No. It's not so much about the end of the road, as it is explaining the bricks in the path along the way.
If by would it matter you meant would it change the direction of out active investigations, sure. but us living inside a simulation implies the existence of a world that is outside of said simulation, thus only adding another bump in the road before attempting to investigate life. Also the script could be altered or deleted, making it impossible for us to maintain steady progress or escape.
If by would it matter you meant would it leave us in a state of rampant nihilism, we already are, to a point.
