Essentially the idea is to make a highly infectious virus that spreads quickly, using humans and also reservoir animals. Nature already has examples of this.
In addition the virus contains a key, so that if the key matches a sufficiently large features, the virus causes a lethal effect, but only after sufficient incubation time.
Such key features can be anything, from hair/skin/eye colour to less visible things such as red/green colour blindness (very common in Iraq), lack of Neanderthal genes (Africa south of Sahara) etc. It is a hard problem not to make an own goal. The US in particular cannot avoid huge own collateral damages.
