Help me figure out this protocol, i'm trying to replicate this experiment i found on a peper but instructions are unclear.
"One gram of dried baker’s yeast S. serevisiae
was added to 100 ml of PBS, and incubated at room temperature for 18 h. After autoclaving and washing three times with PBS,
the yeast cell suspension (2×108 cells/ml) was used..."
1) Why do they autoclave the yeast suspension? I don't see why it's necessary to kill it since they just need to test a protein agglutination properties to sugars present on the surface of S. cerevisiae. note that the exact function of this protein in nature is exactly to agglutinate microbiota so that they can be easily disposed.
2) won't autoclaving yeast fuck up its sugars (Maillard reaction)?
i can't ask this on adv, they don't know shit and it's full of dumb girls
