We are producing some recombinant protein placed on pet-3a vector in bl21 e coli. It had been suggested to us to induce production with IPTG, and also pet manual says we should "for pET constructions carrying "plain" t7 promoter, a final concentration of 0.4 mM iptg is recommended for full induction", but it doesn't make sense to me because the pet-3a vector has no lac operon. What does then IPTG induce?
