Hanzo, Doomfist and Lucio from existing characters all could benefit from more lore and a relationship/gay reveal could be part of that.
With that said, people tend to forget that there already is a gay/bi male character in the game lore, briefly mentioned in the Sombra ARG.
The leaked email states:

Silvia and Rodrigo seem to be a couple that dined with Guillermo, and whoever this Miguel is. In this context, I think it's fair to assume Miguel is Guillermo's significant other, but who can say.
With LumeriCo being connected to several people and factions (Sombra, Vishkar, Los Muertos, Volskaya Industries), it makes sense that we'll get more on LumeriCo and the ongoing storyline with Sombra, Katya and Zarya that even LumeriCo is tangled in, and Miguel could be a new character that'll progress that storyline one way or another. Problem is we already have Sombra who is of Mexican heritage, but they could just make Miguel a Spaniard, which we don't have yet. We don't have a chill, suave older guy either, all the old farts in the game are obnoxious or cranky.
So for me, it's probably Hanzo, Doomfist, Lucio, Miguel or a completely new character.
