The ratio of C13 to C12 is different for modern organisms than it is for ancient organic material so when fossil fuels are burned it causes a shift in the current atmospheric C ratios that can't be explained by volcanic C13/C12 ratios.
That's not the only means we have of determining CO2 is anthropogenic: my personal favorite for its simplicity is just that we have records of how much fossil fuels and of what kinds humans have been emitting. You can make a carbon budget for the world and take out all the natural sources to see that anthropogenic CO2 is necessary to account for the measured rise.
