Either I am being trolled with sterile broth or a very tiny demon is secretly removing my bacteria. Or I just suck at microbiology.

Are you giving the loop enough time to cool before putting it into your broth? Are you shaking the broth enough to distribute the bacteria? Are you getting a decently saturated swab? Are you heat fixing on the wrong side or for too long?

Actually our lab is fucked so we have no gas at the moment and hence no heat fixing or heating the loops (using disposable loops). I think my shaking is fine. Most other people had decent visuals which drives me up the wall

Are you just using slide covers with no stains or anything? I would say just do a larger culture scoop when you're mixing. What are you guys examining?
