No. At no point did Marco state or hint at being trans or identifying as anything other than male.
Sure the Star vs crew would have loved to have changed that though in the ep with the doll in the OP, Marco pulled a string on its back and it said "Dismantle the Patriarchy".

He stopped wearing the dress in season four. He only ever wore it when either Miss Heinous or St Olgas were involved. Once Heinous reverted back to Meteora Marco never wore the dress again.

Marco kinda disliked the dress, next time he was forced to dress like a princess he was all "can I keep the dress?" out of nowhere... from that point onwards everyone knew cotugno was behind that shit
